產品簡介:胰酶細胞消化液(Trypsin-EDTA Solution)含 0.25%胰酶(Trypsin)和 0.02%EDTA,不含Ca2+和 Mg2+。該消化液已用 0.22 μm 濾膜過濾除菌,可以直接用于培養細胞的消化,或者一些組織的消化。本胰酶細胞消化液具有方便快速的特點,通常室溫消化 1 分鐘左右就可以消化下大多數貼壁細胞。使用說明:貼壁細胞的消化:1、吸去培養液,用無菌的 PBS、Hanks 液或無血清培養液洗滌細胞一次,以去除殘余的血清。2、加入少量胰酶細胞消化液,略蓋過細胞即可,室溫放置 30 秒至 2 分鐘。不同的細胞消化時間有所不同。3、顯微鏡下觀察,細胞明顯收縮,并且肉眼觀察培養器皿底部發現細胞的形態發生明顯的變化,或者用槍吹打細胞發現細胞剛好可以被吹打下來;此時吸除胰酶細胞消化液;加入含血清的完全細胞培養液,吹打下細胞,即可直接用于后續實驗。4、如果發現消化不足,則加入胰酶細胞消化液重新消化。5、如果發現細胞消化時間過長,未及吹打細胞,細胞已經有部分直接從培養器皿底部脫落,直接用胰酶細胞培養液把細胞全部吹打下來。1000-2000g 離心 1 分鐘,沉淀細胞,盡量去除胰酶細胞消化液后,加入含血清的完全培養液重新懸浮細胞,即可用于后續實驗。 組織的消化:不同的組織需要消化的時間相差很大,通常以消化后可以充分打散組織為宜。注意事項:1、 在使用胰酶細胞消化液的過程中要特別注意避免消化液被細菌污染。2、 胰酶細胞消化液消化細胞時間不宜過長,否則細胞鋪板后生長狀況會較差。保存條件:4℃保存;長期貯存于-20℃中;-20℃貯存下保存一年。
